gram stain kit (4-step) principle
gram staining method was developed in 1884 by danish physician gram. gram-positive bacteria possess thick cell walls with abundant and tightly cross-linked peptidoglycan layers. after gram staining, the crystal violet-iodine complex can be firmly retained within cell walls and hardly removed by decolorizer, showing purple color. the gram stain kit is developed based on this principle. gram staining is a widely applied differential staining technique (counterstain method) in bacteriology. it can be used for specimen smears and colony smears. staining creates obvious contrast between bacteria and background, allowing clear observation of bacterial morphology, arrangement and structural features for clinical classification and identification. staining results divide bacteria into two categories: gram-positive bacteria (purple) and gram-negative bacteria (red). this rapid gram stain kit shortens staining time. the whole staining process only takes about 1 minute with nearly no false-negative results, making it an ideal choice for microbiology laboratories in clinical testing.

storage conditions and shelf life
unopened staining solutions have a shelf life of 24 months. once opened within the valid period, the solutions should be used up within 6 months. tightly close the bottle cap after each use to prevent volatilization and deterioration. store the reagents in a well ventilated environment at 5Celsius~30Celsius, with relative humidity not more than 80% and no corrosive gas.
specimen requirements
prepare smears carefully to achieve thin and uniform layers. air-dry prepared slides naturally. if heat fixation is adopted, avoid excessive temperature; the back of slide should feel warm but not hot against the back of hand. overheating may change bacterial morphology and affect observation results. |
